2019 nCoV is an enveloped, single, and positive-stranded RNA virus. The virus encodes four major structural proteins that have played important roles in causing the infection (*): Spike protein (S protein), nucleocapsid protein (N protein), membrane protein (M protein), and small envelope protein (E protein) (Fig.1).
S protein is a large type I transmembrane protein, composed of two sub-units S1 and S2. S protein is essential for both host specificity and viral infectivity.
For viral entry, the S1 sub-unit binds via its RBD (Receptor Binding Domain) to the cellular receptor angiotensin-converting enzyme 2 (ACE2) and after cleavage by proteases (Furin and TMPRSS2), the active S2 sub-unit fuses to the viral membranes with the host cell membrane. The spike protein is reported to induce the neutralizing-antibody and T-cell responses, as well as protective immunity. The CoV spike (S) glycoprotein therefore represent the key target for vaccines, therapeutic antibodies, and diagnostics (*).
SARS-Cov-2 can be triggered to fuse at either the plasma membrane or the endosomal membrane, and the route of entry is dependent on protease availability (Fig. 2).
The virus antigens are presented to the immune-system (APCs, macrophage, …) which in turn produce a range of pro- inflammatory "cytokines” (e.g. IL6, IFNα, IFNβ, IFNγ, IFNλ…) and TNF-α (*). In some cases, these cytokines proliferate and activate an uncontrolled and devastating pro-inflammatory response in the host.
