A colorimetric microassay for quantification of Keratinocyte Transglutaminase (TG1) activity

Principe | The TG1-Covtest uses biotinylated K5 peptide (Biotin-pepK5) as the first substrate (amineacceptor/ acyl-donor) and spermine as second substrate (amine-donor/acyl-acceptor). In the presence of Tgase-1 (suspected to be contained in the samples), spermine is incorporated into the gamma carboxamide of the glutaminyl residue of the biotin-pepK5. The quantity of biotin incorporated is related to the Tgase-1 activity and is revealed by a peroxidase colorimetric reaction. |
