FasL (CD95L / CD178) binds to Fas (CD95), a receptor that transduces the apoptotic signal into cells. It is involved in cytotoxic T cell-mediated apoptosis and in T cell development. The formation of the Fas death-inducing signalling complex (DISC) is the initial step of Fas signalling. Activation of procaspase-8 at the DISC leads to the induction of death receptor (DR)-mediated apoptosis.
Stimulation of Fas has also been reported to trigger non-apoptotic pathways. It has been shown that membrane-bound FasL is essential for the cytotoxic activity, whereas soluble FasL appears to promote autoimmunity and tumorigenesis via the induction of non-apoptotic pathways, in particular NF-κB. FasL also binds to decoy receptor 3 (DcR3).
