Glucocorticoid-induced TNF receptor ligand (GITRL) is preferentially expressed by regulatory T cells and binds to GITR (CD357). It is constitutively expressed and released as soluble form by solid tumors and various hematopoietic malignancies. GITRL causes differentiation of osteoclasts, activation of macrophages, but also alteration of carcinoma and leukemia cells, and influences T cell proliferation as well as TCR-mediated apoptosis.
GITRL is implicated in the development of autoimmune diseases and in the immune response against infectious pathogens and tumors. Besides, GITR serves as a negative regulator for NK cell activation and mediates NF-κB activation via the TRAF2/NIK pathway.
